STON2 variations are involved in synaptic dysfunction and schizophrenia-like behaviors by regulating Syt1 trafficking - EurekAlert
<p>Synaptic dysfunction plays an important role in the pathophysiology of schizophrenia. Ma et al. showed that the C-C (307Pro-851Ala) haplotype of STON2 increases the risk of schizophrenia, and STON2<sup>307Pro851Ala</sup> knockin mice exhibited schizophrenia-like behaviors and effects on clathrin-mediated endocytosis (CME) in excitatory synapses. Pharmacogenomic data revealed that patients with the C-C (307Pro-851Ala) haplotype respond better to haloperidol, and haloperidol administration restored the schizophrenia-related behaviors of STON2<sup>307Pro851Ala</sup> knockin mice through the regulation of CME. Their findings suggest that STON2 genotyping could be used for personalized therapy for schizophrenia patients.</p>
